Behold, the artificial jellyfish: Researchers create moving model, using silicone polymer and heart muscle cells

ScienceDaily (July 22, 2012) ? Using recent advances in marine biomechanics, materials science, and tissue engineering, a team of researchers at Harvard University and the California Institute of Technology (Caltech) have turned inanimate silicone and living cardiac muscle cells into a freely swimming "jellyfish."

The finding serves as a proof of concept for reverse engineering a variety of muscular organs and simple life forms. It also suggests a broader definition of what counts as synthetic life in an emerging field that has primarily focused on replicating life's building blocks.

The researchers' method for building the tissue-engineered jellyfish, dubbed "Medusoid," was published in a Nature Biotechnology paper on July 22.

An expert in cell- and tissue-powered actuators, coauthor Kevin Kit Parker has previously demonstrated bioengineered constructs that can grip, pump, and even walk. The inspiration to raise the bar and mimic a jellyfish came out of his own frustration with the state of the cardiac field.

Similar to the way a human heart moves blood throughout the body, jellyfish propel themselves through the water by pumping. In figuring out how to take apart and then rebuild the primary motor function of a jellyfish, the aim was to gain new insights into how such pumps really worked.

"It occurred to me in 2007 that we might have failed to understand the fundamental laws of muscular pumps," says Parker, Tarr Family Professor of Bioengineering and Applied Physics at the Harvard School of Engineering and Applied Sciences (SEAS) and a Core Faculty Member at the Wyss Institute for Biologically Inspired Engineering at Harvard. "I started looking at marine organisms that pump to survive. Then I saw a jellyfish at the New England Aquarium and I immediately noted both similarities and differences between how the jellyfish and the human heart pump."

To build the Medusoid, Parker collaborated with Janna Nawroth, a doctoral student in biology at Caltech and lead author of the study, who performed the work as a visiting researcher in Parker's lab. They also worked with Nawroth's adviser, John Dabiri, a professor of aeronautics and bioengineering at Caltech, who is an expert in biological propulsion.

"A big goal of our study was to advance tissue engineering," says Nawroth. "In many ways, it is still a very qualitative art, with people trying to copy a tissue or organ just based on what they think is important or what they see as the major components -- without necessarily understanding if those components are relevant to the desired function or without analyzing first how different materials could be used."

It turned out that jellyfish, believed to be the oldest multi-organ animals in the world, were an ideal subject, as they use muscles to pump their way through water, and their basic morphology is similar to that of a beating human heart.

To reverse engineer a medusa jellyfish, the investigators used analysis tools borrowed from the fields of law enforcement biometrics and crystallography to make maps of the alignment of subcellular protein networks within all of the muscle cells within the animal. They then conducted studies to understand the electrophysiological triggering of jellyfish propulsion and the biomechanics of the propulsive stroke itself.

Based on such understanding, it turned out that a sheet of cultured rat heart muscle tissue that would contract when electrically stimulated in a liquid environment was the perfect raw material to create an ersatz jellyfish. The team then incorporated a silicone polymer that fashions the body of the artificial creature into a thin membrane that resembles a small jellyfish, with eight arm-like appendages.

Using the same analysis tools, the investigators were able to quantitatively match the subcellular, cellular, and supracellular architecture of the jellyfish musculature with the rat heart muscle cells.

The artificial construct was placed in container of ocean-like salt water and shocked into swimming with synchronized muscle contractions that mimic those of real jellyfish. (In fact, the muscle cells started to contract a bit on their own even before the electrical current was applied.)

"I was surprised that with relatively few components -- a silicone base and cells that we arranged -- we were able to reproduce some pretty complex swimming and feeding behaviors that you see in biological jellyfish," says Dabiri.

Their design strategy, they say, will be broadly applicable to the reverse engineering of muscular organs in humans.

"As engineers, we are very comfortable with building things out of steel, copper, concrete," says Parker. "I think of cells as another kind of building substrate, but we need rigorous quantitative design specs to move tissue engineering to a reproducible type of engineering. The jellyfish provides a design algorithm for reverse engineering an organ's function and developing quantitative design and performance specifications. We can complete the full exercise of the engineer's design process: design, build, and test."

In addition to advancing the field of tissue engineering, Parker adds that he took on the challenge of building a creature to challenge the traditional view of synthetic biology which is "focused on genetic manipulations of cells." Instead of building just a cell, he sought to "build a beast."

Looking forward, the researchers aim to further evolve the artificial jellyfish, allowing it to turn and move in a particular direction, and even incorporating a simple "brain" so it can respond to its environment and replicate more advanced behaviors like heading toward a light source and seeking energy or food.

Along with Parker, Nawroth, and Dabiri, contributors to the study included Hyungsuk Lee, Adam W. Feinberg, Crystal M. Ripplinger, Megan L. McCain, and Anna Grosberg, all at Harvard.

(AP) ? "The Dark Knight Rises" star Christian Bale says his heart goes out to the victims of the Colorado shootings at a screening of the Batman film.

In a statement released Saturday, Bale says, "Words cannot express the horror that I feel." He says: "I cannot begin to truly understand the pain and grief of the victims and their loved ones, but my heart goes out to them."

Bale plays the caped crusader in the film.

Meanwhile, Sony, Fox, Disney, Universal, Fox and Lionsgate said Saturday that they are joining "Dark Knight Rises" distributor Warner Bros. in withholding their box-office numbers for the weekend.

The shootings at the midnight Friday showing in Aurora, Colo., killed 12 and wounded 58. A suspect is in custody.

In Israel, Romney to provide contrast with Obama

WASHINGTON (Reuters) - Republican presidential candidate Mitt Romney will soon do something Barack Obama has yet to do as president - visit U.S. ally Israel, where he will try to present himself to voters back home as a credible replacement to Obama on the world stage.

In the midst of a presidential campaign that is too close to call, Romney leaves on Wednesday for a week-long trip to attend the opening ceremonies of the Olympics in London and visit Israel and Poland.

The aim is to give the former Massachusetts governor some foreign policy credentials and let Americans get a glimpse of him in action overseas as they decide whether to vote for him in the November 6 general election to replace Obama.

Aides say the trip is a "listen and learn" tour with no policy pronouncements. It is a lower key version of Obama's own 2008 trip abroad in which he spoke to a huge throng in Berlin in the heat of that presidential campaign and declared, "The walls between old allies on either side of the Atlantic cannot stand."

Romney's visit to the Olympics will allow him to wave the American flag and play up a central piece of his resume - his work to salvage the 2002 Winter Olympics in Salt Lake City.

In London he will meet Prime Minister David Cameron and other officials as well as former Prime Minister Tony Blair to stress the importance of traditional U.S.-British ties. He will also raise some campaign cash from Americans living in Britain.

In Warsaw and Gdansk, Romney will hold talks with Polish officials and former Solidarity leader Lech Walesa and voice support for an ally that has stood as a key bulwark in eastern Europe, a region long dominated by Russia.

"The trip is to demonstrate a clear and resolute stand with nations that share our values," said Romney policy adviser Lanhee Chen.

Israel is the most delicate diplomatic stop for Romney, with Syria in turmoil after a bomb attack in Damascus killed the defense minister, and Israeli tensions with Iran rising after a bus bomb in Bulgaria killed six Israeli tourists and prompted Israel to blame Tehran.

'WALKING BETWEEN THE RAINDROPS'

Romney's visit presents him the opportunity to appeal to both Jewish voters and pro-Israel evangelical voters and contrast himself with the Democratic incumbent Obama, who has a rocky relationship with Prime Minister Benjamin Netanyahu.

But it also offers some risks as Romney, a relative novice in foreign policy, may be asked whether he would go to war to halt Iran's nuclear program, a top concern of Israel, and whether he would get the United States involved in the bloody turmoil in neighboring Syria, where President Bashar al-Assad is under siege from rebels.

Romney has sharply criticized Obama's handling of Iran and adamantly declared he would not allow it to possess a nuclear weapon, which Tehran denies seeking. Romney has said that "ultimately, regime change is what's going to be necessary."

This has left him open to questions over whether he would launch a military operation against Iran at a time when Americans are weary of wars in Afghanistan and Iraq begun by the last Republican president, George W. Bush.

"That's the key consideration," said Martin Indyk, director of foreign policy at the Brookings Institution and author of the book, "Bending History: Barack Obama's foreign policy."

"On the one hand Romney wants to be able to criticize Obama as being weak on Iran, but I don't think he wants to portray himself as eager for a conflict with Iran. So I think he's walking between the raindrops to try to score Obama without creating the impression that he's keen for another war in the Middle East," Indyk said.

Romney has accused Obama of a lack of leadership for failing to gain U.N. passage of a resolution threatening sanctions against Syria.

Lest Romney face criticism for taking the campaign against Obama overseas, the Romney camp stressed that the candidate will not use the trip to make policy pronouncements.

The friendly imagery from Romney's talks with Netanyahu will be critical in providing a contrast with that of Obama and the Israeli leader, who have clashed in the past.

Obama angered the Israelis a year ago when he embraced a goal long sought by the Palestinians, that the state they seek in the occupied West Bank and Gaza Strip should largely be drawn along lines that existed before the 1967 war in which Israel captured those territories, including East Jerusalem.

"The focus of the trip really is about learning, listening ... and it's about continuing to project Governor Romney's strong view that America needs to stand by its allies, particularly allies that are under siege, like Israel," said Romney foreign policy adviser Dan Senor.

Former Secretary of State Condoleezza Rice visited Jerusalem in May and heard complaints from the Israeli leadership about the current state of the U.S.-Israeli relationship, a source familiar with her meetings said.

It has not gone unnoticed there that the Democratic president has not visited Israel, although he has sent some ranking officials there, including Secretary of State Hillary Clinton.

"It's not a trivial issue that Obama has not been to Israel during his presidency," said Michael Goldfarb, a Republican foreign policy expert. "Of course he went as a candidate, but he ran as a pro-Israeli figure and in office has not lived up to his promises."

Romney, who says Obama has proposed Israel adopt "indefensible borders," has known Netanyahu since the 1970s when they both worked briefly at the Boston Consulting Group. Romney also plans to meet Palestinian leaders.

"There's no question that Romney needs to be careful," said Shibley Telhami, a Middle East expert at the University of Maryland. "He needs to come across as a statesman rather than a panderer and be careful not to lock himself into positions that would tie his hands down the road."

While American Jewish voters overwhelmingly support Democrats, Romney's strong pro-Israeli stance could help him cut into Obama's dominance with this voting bloc, which could make a difference in a battleground state like Florida.

Perhaps more importantly, the Israel trip could energize staunchly pro-Israel evangelical conservatives who have been suspicious of Romney - a Mormon who has a history of being moderate - and had searched for an alternative during the Republican primary battle.

Mammography Screening Shows Limited Effect On Breast Cancer ...

Breast cancer mortality statistics in Sweden are consistent with studies that have reported that screening has limited or no impact on breast cancer mortality among women aged 40-69, according to a study published July 17 in the Journal of The National Cancer Institute.
Since 1974, Swedish women aged 40-69 have increasingly been offered mammography screening, with nationwide coverage peaking in 1997. Researchers set out to determine if mortality trends would be reflected accordingly.
In order to determine this, Philippe Autier, M.D., of the International Prevention Research Institute (iPRI) in France and colleagues, looked at data from the Swedish Board of Health and Welfare from 1960-2009 to analyze trends in breast cancer mortality in women aged age 40 and older by the county in which they lived. The researchers compared actual mortality trends with the theoretical outcomes using models in which screening would result in mortality reductions of 10%, 20%, and 30%.
The researchers expected that screening would be associated with a gradual reduction in mortality, especially because Swedish mammography trials and observational studies have suggested that mammography leads to a reduction in breast cancer mortality. In this study, however, they found that breast cancer mortality rates in Swedish women started to decrease in 1972, before the introduction of mammography, and have continued to decline at a rate similar to that in the prescreening period. ?It seems paradoxical that the downward trends in breast cancer mortality in Sweden have evolved practically as if screening had never existed,? they write. ?Swedish breast cancer mortality statistics are consistent with studies that show limited or no impact of screening on mortality from breast cancer.?
The researchers do note certain limitations of their study ? namely, that it was observational, so unable to take into account the potential influence of other breast cancer risk factors such as obesity, which may have masked the effect of screening on mortality. They also write that population mobility may have biased the results.

Nexus S on Vodafone Australia won't be getting Jelly Bean today after all

Continuing this morning's Jelly Bean soap opera, we've just heard that Nexus S owners on Vodafone Australia won't be getting their pioneering update today -- despite the fact that only 24 hours have elapsed since the carrier said the roll-out was on its way. According to the carrier's official blog, the update has been delayed because Android 4.1 "does not meet all Australian regulatory requirements related to emergency calls." Ouch. On the other hand, that sounds eminently fixable.

Baseball in the Time of Cholera screening

On Wednesday, July 18, an important film screening and discussion was held in Congress.? Co-produced by actress Olivia Wilde, and awarded Best Documentary Short at the Tribeca Film Festival, ?Baseball in the Time of Cholera? documents Haiti?s cholera epidemic through the eyes and voice of a young baseball player.? It provides a powerful insight into the tragedy and scandal of this crisis, as well as the hope and resilience of the Haitian people.

The public release of this film coincided with the delivery of a letter, signed by 104 members of the U.S. Congress, to Susan Rice, U.S. Ambassador to the United Nations.? The letter requests that Ambassador Rice urge the world body to act decisively to address Haiti?s cholera crisis.? MCC has joined other Human Rights groups, faith-based organizations and NGOs in commending this letter.???Members of Congress have joined a growing chorus calling for UN authorities to work with Haiti?s government and the Haitian people to confront and ultimately eliminate cholera,? said Rachelle Lyndaker Schlabach, director of MCC US Washington Office.

Anti-tau drug improves cognition, decreases tau tangles in Alzheimer's disease models

ScienceDaily (July 19, 2012) ? While clinical trial results are being released regarding drugs intended to decrease amyloid production -- thought to contribute to decline in Alzheimer's disease -- clinical trials of drugs targeting other disease proteins, such as tau, are in their initial phases.

Penn Medicine research presented July 19 at the 2012 Alzheimer's Association International Conference (AAIC) shows that an anti-tau treatment called epithilone D (EpoD) was effective in preventing and intervening the progress of Alzheimer's disease in animal models, improving neuron function and cognition, as well as decreasing tau pathology.

By targeting tau, the drug aims to stabilize microtubules, which help support and transport of essential nutrients and information between cells. When tau malfunctions, microtubules break and tau accumulates into tangles.

"This drug effectively hits a tau target by correcting tau loss of function, thereby stabilizing microtubules and offsetting the loss of tau due to its formation into neurofibrillary tangles in animal models, which suggests that this could be an important option to mediate tau function in Alzheimer's and other tau-based neurodegenerative diseases," said John Trojanowski, MD, PhD, professor of Pathology and Laboratory Medicine in the Perelman School of Medicine at the University of Pennsylvania. "In addition to drugs targeting amyloid, which may not work in advanced Alzheimer's disease, our hope is that this and other anti-tau drugs can be tested in people with Alzheimer's disease to determine whether stabilizing microtubules damaged by malfunctioning tau protein may improve clinical and pathological outcomes."

The drug, identified through Penn's Center for Neurodegenerative Disease Research (CNDR) Drug Discovery Program, was previously shown to prevent further neurological damage and improve cognitive performance in animal models*. The Penn research team includes senior investigator Bin Zhang, MD, and Kurt Brunden, PhD, director of Drug Discovery at CNDR.

Bristol-Myers Squibb, who developed and owns the rights to the drug, has started enrolling patients into a phase I clinical trial in people with mild Alzheimer's disease.

WASHINGTON (AP) ? For years it seemed hopeless. Now the hunt for a cure for AIDS is back on.

International AIDS specialists on Thursday released what they call a road map for research toward a cure for HIV ? a strategy for global teams of scientists to explore a number of intriguing leads that just might, years from now, pan out.

"Today's the first step," said French Nobel laureate Francoise Barre-Sinoussi, co-discoverer of the HIV virus who also co-chaired development of the strategy.

"No one thinks it's going to be easy," added strategy co-chair Dr. Steven Deeks of the University of California, San Francisco. "Some don't think it's possible."

The announcement came just before the International AIDS Conference begins on Sunday, when more than 20,000 scientists, activists and policymakers gather in the nation's capital with a far different focus: how to dramatically cut the spread of the AIDS virus, what they call "turning the tide" of the epidemic, using some powerful tools already in hand.

Chief among them is getting more of the world's 34 million HIV-infected people on life-saving medications, so they stay healthier and are less likely to infect others. By itself, that is a huge hurdle. Just 8 million of the 15 million treatment-eligible patients in AIDS-ravaged poor regions of the world are getting the drugs.

But Barre-Sinoussi, president-elect of the International AIDS Society, which hosts the conference, said that lifelong treatment, as good as it is, isn't the end-all solution ? and that science finally is showing that a cure "could be a realistic possibility."

The panelists refused to estimate Thursday how much this research would cost. But already, the National Institutes of Health has increased spending on cure-related research, about $56 million last year, according to a report in this week's issue of the journal Nature. Scientists attempting cure research will meet Friday and Saturday, ahead of the AIDS conference, to compare notes.

And the new strategy won praise from Michel Sidibe, executive director of UNAIDS, the Joint United Nations Program on HIV and AIDS.

"The previous generation fought for treatment," he said. "Our generation must fight for a cure."

Today's anti-HIV drugs can tamp down the virus to undetectable levels ? but they don't eradicate it. Instead, tiny amounts of the virus can hide out in different tissues and roar back if medication is stopped.

That means there's no certainty of developing a cure.

"I'm not sure we can, but we're going to try," Dr. Anthony Fauci, director of NIH's National Institute of Allergy and Infectious Diseases, said in a recent interview. "This virus is uncanny in its ability to be able to integrate itself into a cell, as a reservoir, and no matter what we've done so far, we have not been able to eliminate that reservoir."

Yet one person in the world apparently has been cured: Timothy Ray Brown of San Francisco, who in 2006 was living in Berlin when in addition to his HIV, he got leukemia.

Brown underwent a blood stem cell transplant ? what once was a bone marrow transplant ? to treat the cancer. His own immune system was destroyed. And his German transplant surgeon found a donor who was among the 1 percent of whites who have a gene mutation that makes them naturally resistant to HIV ? their cells lack the specific doorway the virus uses to get inside.

It worked. Brown has been off HIV medications for five years and is doing well, Deeks said Thursday.

That dangerous and expensive transplant isn't a practical solution, but it has sparked a variety of research into other possible ways to eradicate HIV. Already, 12 early-stage studies involving small numbers of patients ? fewer than 200 people worldwide ? are under way, the international panel said Thursday. Results to see if any are promising enough to pursue should be out in the next year or two.

The priorities of the new cure research strategy:

?Determine why HIV hibernates and persists.

?Learn why some people are naturally resistant. In addition to that 1 percent of people with the gene mutation, researchers now are studying a small group of patients in France who started medication soon after they were infected and many years later were able to stop the drugs without the virus rebounding.

?Develop and test strategies to make HIV patients more naturally resistant. Already gene therapy studies are under way to knock that HIV doorway out of people's own infection-fighting blood cells.

?Learn where all those secret reservoirs are.

?Develop strategies to attack the reservoirs. One new attempt uses drugs to wake up the dormant HIV so the immune system can spot and attack it, what Deeks called the "shock and kill approach." Last spring, University of North Carolina, Chapel Hill, researchers reported that a drug normally used for lymphoma made some latent HIV rapidly detectable in six patients. Deeks has a similar study under way using an old anti-alcoholism drug.

?Develop good tests to measure these tiny amounts of dormant HIV, crucial to telling if any cure attempts are promising short of taking patients off their regular medication.

To prevent mold and mildew, you should keep your bathroom ventilated well. Such allergens proliferate in warm, wet areas. Hang wet washcloths and towels on bars and turn on the fan after you?ve showered. If you do not have a fan, crack a window to allow fresh air to circulate.

A great way to treat any type of bronchial allergies is to always make sure your body is getting enough fluids. When you are dehydrated, the membranes of your nose and throat can become dry and irritated. Failure to hydrate can also cause the mucosal glands to secrete excess substances.

Keep pets indoors when outdoor pollen is a problem or the humidity is high. Pollen can stick to your pet?s coat and be brought indoors, worsening your allergies.

The western part of the country has seen an upsurge in the popularity of olive trees as decoration. These trees produce a lot of pollen. Educating yourself about this tree can allow you to enjoy it in your surroundings, while still forming a plan of attack against your allergies. A lot of people realize that turning a hose to the tree for a couple of minutes a day will reduce pollen issues.

If you are experiencing seasonal allergies your might want to wash your hair and change your clothes, or even take a shower as you enter your house from the outside. If you don?t, you may make your allergies worse because you will bring allergens in your home.

Children going to school with allergies, may be using medications to handle severe allergic reactions. You may also want to get a formal note from your pediatrician explaining your child?s condition and emergency treatment.
